During the night the tv turns on, showing the stand by screen. We have tried turning off various options but to no avail.

When Sky does a back up during the night it turns the screen on that shows the icons showing the tv and Sky box etc

It's not always HDMI CEC doing it. In fact it should not be doing it as the Sky box doesn't send the signal when it "turns on" after it's nightly reboot. Not having HDMI CEC turned on can be a bit inconvenient.
We had a similar issue when we first got our replacement Panasonic TV a few years ago. Here's the archived post I logged on that very subject:

It's got all the fancy HDMI stuff, but (for legacy device support) it can also detect any change of signal on selected HDMI ports and auto-switch to that port.
With this option turned on the TV was turning on each morning when the Sky box rebooted.
I don't know whether Samsungs have a similar option? Most TVs have so many options now-a-days and of course each manufacturer makes up their own terms so it's hard to match options from manufacturer to manufacturer.
