Do I send both the Q box and multi room box along with the cables back to sky with the courier when the TV is delivered or send through the post office?

Not with the Glass courier. You should receive packaging and instructions for the return of your Sky Q equipment 3 weeks or so after Glass is delivered. If not, or if you prefer not to wait,  you can return them yourself if you have suitable packaging as explained here: https://www.sky.com/help/articles/return-tv-broadband
