Yes it will as the Direct Debit will always be taken even if an uneccessary payment is made so close to the DD date.
In short you don't need to be making manual payments when you have a direct debit in place.

The mistake here is yours by making a manual payment that wasn't needed as you have a direct debit setup. Now you have done this you have two options, leave it place then it will be used against your next bill or it can be refunded and help of which can be arranged via here if you would like ?

As stated manual payments are unnecessary as the T's and C's you’ve agreed to state you should have a direct debit or continuous card payment method set up. You therefore don’t need to make a manual payment unless your services have already been restricted.
You need to let Sky take the planned payment they are expecting, a credit should then appear on your Sky account for the amount you have overpaid. This credit should automatically be used to pay your next months bill so on that bill date no/less money should be taken from your account. I would recommend NOT making any manual payments unless requested to by Sky.
Alternatively once your extra payment is showing as a credit, we can perhaps get you some help in getting it refunded (within the standard 3-5 WORKING days) through staff from the Community Messaging Team on here. Let us know on this thread if you want to do that but please confirm it’s currently showing as a credit.

You need the let the automated collection clear. (The manual payment you've made is a credit against next month's bill)
